I had been brooding over this question for a while and, given the possibility of utilizing this section, I would like to ask: can one enter directly into the two-year program (biennio) or is it mandatory to first go through the three-year degree (triennio)?

Of course it's possible.
The substance is that you have to pass the entrance exam (enrollment is always limited to a certain number) and if you are at 0... you don't get in anyway, degree or no degree.

Well, it wasn't that obvious. If you have a bachelor's degree in Humanities and Philosophy, try enrolling in a master's in engineering... If you manage to do it... 😉
